چکیده مقاله:

Breast cancer is a complex genetic disease that has an average annual incidence of two million people and the second leading cause of death among women all over the world. Micro-RNAs are consistently reported to regulate gene expression in all cancers. The present study, the correlation between the expression of hsa-miR-490-5p and nuclear factor of activated T-cells 5 (NFAT5) in breast cancer were investigated. NFAT5 nuclear accumulation occurs regardless of Wnt/β-catenin activated signaling in a substantial portion of breast cancer. The analysis of prediction target and dual-luciferase reporter assays supported that hsa-miR-490-5p directly targeted NFAT5 and suppressed the expression of NFAT5. In a cross-sectional comparative study, peripheral blood samples were collected from 30 subjects with breast cancer and 30 healthy individuals as a control group. Total mRNA was extracted from peripheral blood mononuclear cells (PBMCs) and cDNA was synthesized to study the NFAT5 and hsa-miR-490-5p gene expression variations by real-time PCR. A significant decrease was observed in gene expression and sera concentration of NFAT5, hsa-miR-490-5p in PBMCs of breast cancer patients. The obtained results indicated that hsa-miR-490-5p acts as oncomir in serum by targeting NFAT5 direct (p

Mir-183 and FOXO1 gene expression changes in peripheral blood mononuclear cells of breast cancer patients

Background and Aim: Today, cancer is considered as a major health problem and affects the health of society. Breast cancer is the second leading cause of cancer death in women after lung cancer. According to epidemiological studies, cancer is the second most common cause of death after cardiovascular disease worldwide and the third leading cause of death after cardiovascular disease and acciden...

MiR-490-5p Functions as an OncomiR in Breast Cancer by Targeting NFATc4

Breast cancer is a serious health problem worldwide in women. MicroRNAs are small non-coding RNAs of 18–25 nucleotides in length that post-transcriptionally modulate gene expression. MiR-490 has been reported as a tumor suppressor and oncomiR microRNA in breast cancer with two separate targets, NFAT and Rho. NFAT is one of the targets for miR-490 but the relationship between hsa</e...

Increased miR-424-5p expression in peripheral blood mononuclear cells from patients with pemphigus

Pemphigus is an autoimmune disease that causes blisters and erosions in the skin and mucous membranes. The development of pemphigus is associated with the imbalance of T‑cell and humoral responses. MicroRNAs (miRNAs) can regulate many cell functions. However, whether miRNA expression is altered in peripheral blood mononuclear cells (PBMCs) during the pathogenesis of pemphigus has not been clari...

Negative Regulation of Semaphorin-3A Expression in Peripheral Blood Mononuclear Cells Using MicroRNA-497-5p

Background: Semaphorin-3A (Sema3A), as a secreted semaphorin, is an immune modulator molecule participating in the pathogenesis of autoimmune diseases. MicroRNAs (miRNAs) modulate the target-gene expression at the post-transcriptional level. It has been proposed that miRNAs may be crucial to the modulation of the function of semaphorins. Previous findings have proven that miR-497-5p is upregula...

Investigation of NK Cell Population in Peripheral Blood and Tumor Lesions of Patients with Breast Cancer

Background: Alteration in peripheral blood lymphocytes (PBLs) is usually investigated to provide an evidence of the host immune responses to tumor antigens. The tumor infiltrating NK cells interact most closely with the tumor cells and more accurately reflect tumor host interactions. Objective: To analyze peripheral blood and tumor associated Natural Killer (NK) cells in patients with breast ca...

Expression of miR-142-5p in Peripheral Blood Mononuclear Cells from Renal Transplant Patients with Chronic Antibody-Mediated Rejection

In renal transplantation, the unresponsiveness of patients undergoing chronic antibody mediated rejection (CAMR) to classical treatment stress on the need for accurate biomarkers to improve its diagnosis. We aim to determine whether microRNA expression patterns may be associated with a diagnosis of CAMR. We performed expression profiling of miRNAs in peripheral blood mononuclear cells (PBMC) of...
